Enzyme, nitrogen, and DNA concentrations in sarcoma 180 in mice treated with 6-mercaptopurine.

چکیده
The concentrations of homogenate-nitrogen, of DNA, and the activities of various nucleotidases, adenosine deaminase, cathepsin and lactic dehydrogenase have been studied in homogenates of Sarcoma 180 from non treated mice, from mice given injec tions of 6-carboxymethylcellulose in saline, and from mice treated with 6-mercaptopurine suspended in saline containing 6-carboxymethylcellulose. Statistically significant increases in the activities of all the nucleotidases, adenosine deaminase, and cathepsin have been observed in tumor homogenates of mice treated with 6-mercaptopurine. Statistically significant decreases in tumor weight, homo genate-nitrogen, DNA, and lactic dehydrogenase have been observed in the same homogenates. The tumor homogenates of the controls receiving 6-carboxymethylcellulose in saline showed only increases in adenosine deaminase and cathepsin which, however, did not reach the activity levels attained in homogenates of mice treated with 6-mercapto purine. The significance of the findings is discussed.

Synergistic Effect of 6-Mercaptopurine and 6-Methylmercaptopurine Ribonucleoside on the Levels of Adenine Nucleotides of Sarcoma 180 Cells1
The synergistic antitumor action of 6-mercaptopurine (6-MP) and 6-methylmercaptopurine ribonucleoside (MMPR) is confirmed with Sarcoma 180 ascites cells. Striking decreases are seen in the adenine nucleotide concentrations in cells incubated for 1 hr with this drug combination, and lesser effects are seen on the guanine nucleotide concentrations. Still greater decreases in adenine nucleotide le...
متن کاملGlycolysis by Sarcoma 180 of mice treated with 6-mercaptopurine. I. The role of various enzymes in lactate production.
Lactate formation from fructose-1,6-diphosphate in homogenates of Sarcoma 180 from mice, given injections intraperitoneally of 6-mercaptopurine, was sharply de creased as compared with that in tumor homogenates of nontreated animals. The activities of aldolase and triosephosphate isomerase, but not of phosphoglyceraldehyde dehydrogenase and phosphoglyceric acid kinase, were significantly lower,...

متن کاملAntagonistic Effects of 6-mercaptopurine and Coenzyme a on Mitochondria and Mitosis in Tissue Culture
The partial mitotic inhibition caused by 6-mercaptopurine in tissue cultures of Crocker mouse sarcoma 180 and embryonic mouse skin is blocked by co-enzyme A. 6-Mercaptopurine and coenzyme A also have opposite effects on mitochondrial morphology. Mitochondria in cells treated with 6-mercaptopurine become thin and fragmented. Coenzyme A blocks this effect, and alone coenzyme A makes for longer an...

Several compounds known to inhibit the growth of Sarcoma 180 were compared for their cytocidal activity in vivo. Standard inocula from donor mice which were treated with massive doses of the drugs were transplanted at 4 and 24 hours after treatment into untreated recipient animals. Viability was evaluated by cell counts and survival time. The effectiveness of triethylenethiophosphoramide and ac...
